Research Interests


Defects in DNA replication have long been suggested to have a causative role in human cancer, however, specific examples have been elusive. This situation has changed recently with the finding that germline exonuclease domain mutations (EDMs) in human replicative polymerases are associated with a colorectal cancer risk, and somatic mutations in polymerases have also been found to be frequent in endometrial cancer. Our aim is to understand how defects in replicative DNA polymerases found in tumours or cancer predisposition syndromes contribute to tumour development and genome instability. My major interest is to model the human DNA polymerase epsilon EDMs in fission yeast, which allows genetic screening, and thus to focus attention on those variants with a significant mutator phenotype. In the long term it may be possible to design drug therapies to target cells expressing mutant polymerases.
